■Protocol①Set LeukoCatchII into a 50 mL centrifugetube, and apply 10 mL of blood.(5 mL whole blood + 5 mL PBS(-))②Filtering starts with natural dropping force.Filtering time is approx. 10 min.③Leuko Filter traps leukocytes.Consequently, leukocytes are almostremoved from the filtrate.⑥Pool the 5 mL of culture medium intoa φ60 mm dish.Put Leuko Filter in the culture medium.⑦Put the dish in the CO2 incubator(37 degree Celsius)⑧After 24 hours, it can collect about 60%leukocytes from whole blood withoutcentrifugation.④Add 2 mL of PBS(-) in order to washthe Leuko Filter trapped leukocytes.Repeat 1 time.⑤Pick up Leuko Filter with a pair of tweezers and remove Leuko Filter.Pay attention NOT to reverse the face of Leuko Filter.Filtrate50 mLCentrifugeTubeLeukoCatchⅡ*figureCollect Leukocyteswhich are trappedby filer of product.Convex side is upper sideAfter filtration of sample,a lot of leukocytes are attached toconvex side(upper side).There are concave side and convex sidein Lueko Filter.Please match upper side of filter to the bottom of dish.Concave side isback sideLeuko Filter is dentedin center of the back side.Convex sideLeukofilterRemoveLeukocytesCollectLeukocytes66 Blood Research Products Blood Research Products Leuko Catch ⅡLeuko Catch Ⅱ
